Possibility of dopaminergic neurons number regulation in the hepatocarcinomas prevention. Laboratory Animals for Science. 2021; 2. https://doi.org/10.29296/2618723X-2021-02-06
The aim of the work was to study the possibility of regulating the number of dopaminergic neurons in high-cancer CBA mice-males (CBA/LacY subline) during spontaneous hepatocancerogenesis. The mice were prophylactically treated with a standardized phytoadaptogene complex (PAC) consisting components of 40 water-alcohol plant extracts including adaptogens Panax ginseng, Schisandra chinensis, Rhodiola rosea as well as compounds of a phenolic nature. PAC has antimutagenic, antioxidant, immunomodulating properties. Experimental animals received 10% PAC solution with drinking water during the first month of postnatal ontogenesis including the period (5-15 days of life) corresponding to the termination of normal liver tissue differentiation. The level of dopaminergic neurons was determined by the immunohistochemical method according to the number of tyrosinehydroxylase-positive cells in the midbrain. The level of young neurons was determined by the immunohistochemical method according to the number of Ki67-positive cells in the subgranular layer of the hippocampal formation dentate fascia. The use of PAC in early postnatal ontogenesis (as a preventive application) promotes the preservation of the dopaminergic neurons high number in the brain during the late ontogenesis of males CBA correlating with a decrease in the incidence of tumour formation and an increase of the animals lifespan. The prevention of dopaminergic neurones loss in this study follows the previous data in C57BL mice with induced parkinsonism. PAC prevented the death of brain dopaminergic neurones by the type of apoptosis suppressing the level of caspase
